The level of contamination by M. bovis was estimated by digestion with an alpha-complementation group h-determinant-specific restriction endonuclease and Southern analysis. The mycobacterial reference strain H37Rv was used as a control. Thirty-five BCG samples (29.6%) contained a single variant at the two loci and 68 samples (57.6%) contained two variants, with 3 samples (2.6%) containing three variants. The co-existence of Ag85A-variant and beta-variant was found in 12.6% of the samples. A high level of contamination by M. bovis was detected in several samples, including those that were negative by PCR analysis. PCR allows the detection of BCG variants at Ag85A and beta-crystallin loci with a specificity and sensitivity comparable to the use of biochemical tests. This PCR test could be an easy, fast, and useful method for identifying BCG variants within the region.1.
